Kinase Profiling Service

LC Sciences offers a comprehensive kinase analysis service using high-density protein kinase substrate (PKS) peptide microarrays built on our proprietary PepArray™ microfluidic technology for proteome-wide kinase profiling, quantitative assessment of kinase activities, and drug discovery and development research.

Kinases are essential regulators of cell signaling pathways, transferring the terminal phosphate group from ATP to the serine, threonine, or tyrosine residues of target proteins. Using custom-synthesized, addressable peptide microarrays (PepArray™) containing specific kinase substrates on a custom-synthesized, addressable LC Sciences' peptide microarray (PepArray™), we can quantitatively measure kinase activity across a wide range of signaling networks.

Quantitative Results

All assays use universal, highly phosphate-specific fluorescent dye staining. Well-designed negative and positive control references ensure reliable and quantifiable results. In situ–synthesized PKS peptide microarrays, using LC Sciences’ PepArray™ microfluidic technology, deliver more uniform spots and provide a more reliable readout of assay signals. Multiple replicates of each peptide sequence also contributes to data confidence.


High-Throughput Microarray Format


Traditional kinase assays focus on only a single or a few peptide or protein substrates.  Profiling kinases on a peptide microarray offers the opportunity to study thousands of specific substrates (up to thousands of peptides per chip) in a single experiment.  A single experiment using our protein kinase substrate (PKS) peptide microarray is equivalent to 41 experiments per 96-well plate. Custom Microarray Content


Customize one of our standard microarrays or design your own PKS peptide microarray. We can synthesize a fully custom microarray designed specifically for your experiment. Our standard format includes 4,000 customer-defined peptide sequences per chip, but arrays with longer peptides and higher densities available. Ask us for assistance with the design of your PKS peptide microarray.

Synthesis of a standard content or custom-designed protein kinase substrate (PKS) peptide microarray on a PepArray™ microfluidics chip


Pre-staining of each chip to evaluate synthesis quality and establish a baseline fluorescence signal before applying the kinase sample


Quality testing of the received kinase sample


On-chip enzymatic reactions


Selective staining of phosphate groups on phosphopeptides using a highly specific fluorescent dye


Array imaging and data extraction


Data analysis – Includes basic data processing (background subtraction, control- and reference-guided signal processing, list of detected signals, data averaging, results summarized in a report) is included with our array service.  Optional in-depth analysis based on your specific application is available.
